Messkolben haben keine Skalierung. Es muss also immer eine vorgegebene Menge an Lösung angesetzt werden. Messkolben gehören neben Vollpipetten und Büretten zu den genausten Volumenmessgeräten.
Am engen Hals haben Messkolben einen einzigen Eichstrich, der als Ringmarke ganz um den Hals gezogen ist. Damit werden Ablesefehler vermieden.
Durch den schmalen Hals kann das eingefüllte Volumen sehr genau abgelesen werden. Das Füllvolumen ist korrekt, wenn der untere Meniskus (Wölbung) der Flüssigkeit auf der Ringmarke liegt. Eine maximale Genauigkeit ist nur gegeben, wenn keine Gasblasen in der eingefüllten Flüssigkeit vorhanden sind. Durch leichtes Schwenken des Messkolben können eventuelle Gasblasen nach oben entweichen.
Die Messkolben aus Borosilikatglas sind auf Einguss geeicht. Das bedeutet, dass die eingefüllte Lösung, bzw. Flüssigkeit der Volumenangabe entspricht. Wird die eingefüllte Menge jedoch in ein anderes Gefäss gefüllt, ist das Flüssigkeitsvolumen geringer, da ein kleiner Teil im Messkolben zurückbleibt.
Das Erhitzen von Messkolben kann zu Verlust der Messgenauigkeit führen.
